1.1. Genealogy of the Phrase "Playing God"

The phrase "playing God" is no neutral observation of a fact. It is the condensation of a tripartite narrative structure spanning cultures, epochs, and traditions: the existence of a sacred boundary, the transgression of that boundary by an illegitimate agent, and the inevitable punishment that follows. Comprehending the phrase demands beginning with the narratives nourishing it, because the objection derives its force not from an explicit logical argument, but from mythological resonance, the feeling that certain things "simply ought not to be done" because they violate something deeply inscribed in the order of things.

The Promethean narrative is the first incarnation of the objection. Prometheus, a Titan living between gods and humans, takes pity on primitive humanity living in cold and darkness. He steals divine fire, the exclusive property of the Olympians, and offers it to humans. The theft is a metonymy for a greater good: knowledge, technique, the power of transformation. Fire is that which permits humans to emerge from the wild state, domesticate food, construct civilisation.

Yet the theft possesses an inexorable consequence: Zeus punishes Prometheus with eternal torment. Chained to a rock in the Caucasus, a divine eagle devours his liver daily. The liver regenerates at night and the torment recommences. Prometheanism, the aspiration to attain powers that seem to belong to a separate domain, is punished not as an occasional act, but as a foundational sin. The punishment is structural: it is no incident that can be avoided through better conduct; it is the ineluctable consequence of transgressing a boundary that ought not to have been crossed.

The myth encodes a lesson: there are privileges belonging to the gods. The human who steals them is not heroic, he is a transgressor. Technique, consequently, is not applied neutrality; it is the theft of divine property. And any technique advancing beyond certain limits, touching domains presumed sacred, is Prometheanism, exposure to chastisement. The synthesis of life would be the supreme theft: the appropriation of the very power to create, which is the ultimate divine privilege. The fear that the phrase "playing God" evokes is not fear of technical failure. It is fear of cosmological transgression, of divine chastisement.

The narrative of the Golem emerges in the medieval Kabbalistic tradition. The Golem is a clay creature animated by a rabbinic sage through the sacred combination of letters of the name of God (Shem). The artificial being exists and obeys its creator until the creator, weary or repentant, erases a letter, and the word dies, the Golem returning to inert clay.

The structure of the myth differs from that of Prometheus. It is not the theft of divine property. It is an attempted appropriation of the power of creation through a simulacrum. The rabbi is no true creator, he is a magician imitating genuine creation. True creation, which would be the unique privilege of God, remains inaccessible. What the magician does is simulation: the manipulation of divine names to produce the appearance of life. The Golem is feigned life, life that can be erased because it was never properly alive, it was merely an artefact manipulated through sacred names.

The lesson is subtle yet crucial: human creation of life is always a falsification of true creation. The very act of creating fails because the human lacks the divine authority rendering creation legitimate. Consequence: artificial life, at best, is imitation; at worst, it is the profanation of names and powers that ought to remain inaccessible. The Golem remains an obedient servant, until the creator repents and dissolves the creation. This encodes a truth: artificial creation is always dependent upon the will of the creator. It lacks genuine autonomy. Hence: synthetic life will never be truly alive, it will always be a manipulated artefact.

Mary Shelley offers a third narrative, and the most complex. Victor Frankenstein achieves what the medieval magician could not: he creates a creature that is truly alive, sensitive, intelligent, capable of language and emotion. It is no simulacrum; it is an authentic being. But the tragedy that follows derives not from creation itself; it derives from abandonment.

As soon as the creature opens its eyes and Victor experiences aesthetic horror, disgust at the appearance of the being he created, , he flees. He abandons the newborn creature, alone, without guidance, without community. The creature, intelligent and endowed with emotional capacity, suffers radical rejection. It seeks recognition from the creator who refuses it. It undertakes desperate searches for a community that rejects it. It is educated merely by books found accidentally. It experiences pure existential solitude: it is the sole being of its species, created by accident, rejected by the only one who could comprehend it as its creator. The tragedy culminates in violence, the creature kills those whom Victor loves, in response to originative abandonment.

Yet Shelley is clear: the crime is not creation. The crime is creation without responsibility. Victor is condemned not for having synthesised life; he is condemned for having created and denied responsibility. Had he recognised the creature as his created being, had he offered care, education, community, , the story would have been different. This is a lesson radically different from that of Prometheus or the Golem. It is not "do not create". It is "if you create, you are responsible". Rejection is a greater crime than creation.

These three narratives, Prometheus, Golem, Frankenstein, encode variations upon a single theme: human creation of life transgresses a boundary that ought not to be crossed. But the type of transgression differs in each case. Prometheus is the theft of divine property. Golem is a simulacrum falsifying true creation. Frankenstein is the abandonment of responsibility. Despite these differences, all three share an assumption: there is a separate, sacred domain belonging exclusively to the gods or to untouchable nature. The human touching that domain violates a law prior to any human law.

In the twentieth century, the phrase "playing God" consolidated itself as an automatic objection. Cloning? "Playing God". GMOs? "Playing God". Synthesis of artificial life? "Playing God". The phrase functions as a pre-deliberative refusal mechanism. It enters into no technical discussion concerning viability, risks, or responsibility. It refuses, ex ante, the very right to investigate. It transforms a technical question into a theological question, and the theological question is positioned as undecidable and prior to any deliberation.

Consequence: the synthesis of life becomes unthinkable. Not because technique is impossible, Syns (cells with completely synthetic genomes) have demonstrated technical viability since 2010. It becomes unthinkable because the theological objection paralyses deliberation before it begins. There is an authority prior to science, ethics, and politics, an authority saying "no" before the question can be posed.

The "playing God" objection follows an implicit syllogistic form: (1) There is a sacred domain belonging to the untouchable divine or natural sphere, fundamentally forbidden to human action because it is property transcending the material order. (2) The synthesis of artificial life is an operation upon this sacred domain, an attempt to wrest from the divine the supreme power to create. (3) Hence, the synthesis of life is a transgression, absolutely forbidden, not for technical or prudential reasons, but for a theological reason prior to any calculation of consequences.

The force of the objection rests entirely upon the truth of both premises. If either premise fails, the entire structure collapses. This chapter proposes: within an immanent ontological framework, that which modern materialism established, , both premises are unfounded. There is no separate sacred domain. And if there is none, the synthesis of life is no transgression of divine property, it is the continuation of a process that living matter has always executed, now including technical intervention as one of its modes of reorganisation.

1.2. The Three Theological Assumptions Deconstructed

The phrase "playing God" is no simple insult. It presupposes at least three conceptual structures whose validity can be tested against the immanent ontology established by contemporary materialism. These three structures, if true, would legitimise the objection. If they fall, the objection reveals itself as linguistic inertia, the survival of a vocabulary whose anchoring was removed. The task of this section is to explicitate and test each assumption.

Classical theology presupposes: there exists a being separate from the world, eternal and immutable, who created the world out of absolute nothing (creatio ex nihilo) and governs it according to divine will. This creator is transcendent, outside the world, not part of it, suffering not the limitations and changes affecting material entities. Transcendence is fundamental: without separation from the world, the creator would not be truly a creator, would be merely part of what was created.

Consequence: life is the property of the creator. Life is no natural property of matter. Matter by itself would be inert, dead, incapable of self-organisation without divine intervention. Life is a conferred breath (pneuma, Aristotelian soul, medieval anima); a gift coming from outside. Hence, the human, a creature made of matter, cannot arrogate the power to create life as does the transcendent creator. The creation of life by a creature is the usurpation of divine privilege.

Ontological materialism removes the need for transcendence. Spinoza formulates this clearly: Deus sive Natura, God is the name for the totality of Nature, not a separate agent. There is no creator outside the world governing it. There is merely Nature, continuous material processes determining one another mutually according to their own laws. These processes possess no exterior cause. Nature is causa sui, self-caused. This dissolves completely the theology of transcendent creation.

The modern world functions, ontologically, without the need for transcendence. Laws of physics describe themselves completely via immanent causes, material processes emerging from prior conditions according to deterministic or stochastic laws, but always within continuous immanence. There is no appeal to an external transcendent cause, no divine decision at an original moment. There is merely matter reorganising under constraints it faces. If this framework is correct, , then the transcendent creator is no necessary hypothesis. It is a construct of tradition persisting through historical inertia, not logical necessity.

Without a transcendent creator, the authority forbidding human creation of life falls. If life is no divine property because there is no separate divine, if life is merely a material process satisfying organisational functions, , then life is, in principle, fabricable. Not because the human is elevated to divine status, but because the divine does not exist as a separate entity monopolising creation. There is merely immanence where the human is a material agent among other material agents, capable of reorganising matter because made of matter.

The second assumption maintains: life is no natural property of matter; it is a conferred property, given as a gift. Inert matter by itself, "inert" in the pre-modern sense of incapable of self-movement, organisation, or spontaneous generation. Life would arrive from outside, insufflated, a supernatural gift.

This possessed coherence in an era when life and death seemed absolute, there was a mysterious force separating the living from the dead, and the human understood not what it was. Inert matter described by classical physics (Newton, Descartes) was res extensa, pure extension, lacking capacity for self-organisation. How could matter so inert generate something alive, sensitive, reproductive? It seemed necessary to postulate an additional force, soul, pneuma, élan vital, coming from outside and transforming inertia into life.

Contemporary materialism inverted the image. Life is no property conferred from outside. It is a property emerging when matter reaches a certain degree of organisational complexity. Matter is not inert in the absolute sense. It is capable of self-organisation under certain conditions. When molecules combine in certain ways (with capacity for replication, metabolism, variation), the regime we call "life" emerges. It is no magical property; it is a physico-chemical regime satisfying certain functional criteria.

Miller-Urey (1952) demonstrated that pre-biotic molecules can form abiotically in an environment resembling primitive Earth. The origin of self-replicators (RNA world) is now understood as a chemical process, not a supernatural intervention. Life does not arrive from outside, it emerges when matter, under specific constraints, organises itself in a new mode.

If life is an emergent property of matter organised in a certain mode, then life is no exclusive property of "true nature" or a "divine creator". It is a property that matter can manifest when reorganised. Hence: if the human manages to reorganise matter such that it satisfies criteria of metabolism, reproduction, and variation, , he will have created genuine life. It will be no falsification or simulacrum; it will be the realisation of a regime that matter can manifest.

The third assumption maintains: there exists a sphere forbidden to the human because that sphere is interdicted by a law transcending human power. Sacred/profane is a real ontological difference, inscribed in the very order of things. Some operations "can" be performed because they belong to the profane, the accessible domain. Others "cannot" because they belong to the sacred, a domain forbidden by its very nature.

This assumption is subtle because it depends not necessarily upon belief in a personal God. It can appear simply as "respect for nature" or "recognition of limits". But underneath there is always the idea: there is something intrinsic in certain domains rendering them "sacred", conferring upon them the property of being untouchable. If life is sacred, however, touching life is profanation.

Sacred and profane are no intrinsic properties of things; they are cultural constructions, attitudes and practices societies adopt toward certain objects or processes. What is sacred varies across epochs, cultures, traditions. In a Greek polytheistic context, gods were sacred, yet immanent, inhabited the world, competed with humans, reproduced. In a Judeo-Christian monotheistic context, only God is sacred and separate. In modern secularism, nothing is sacred because there is no transcendence, merely things society chooses to treat with special care.

This shows: sacred is no ontological quality; it is a cultural attribution. Nothing is sacred "in itself". Something becomes sacred because society, tradition, or religion declares it sacred and confers upon it structured respect, interdict, or cult.

If sacred/profane is a cultural construction and not an ontological property, then "life is sacred" means not "life possesses an intrinsic property rendering it untouchable". It means "our tradition treated life as worthy of special care and respect". This is morally important, care for life is a value worth preserving. But it is different from saying "life is sacred, therefore cannot be touched". It is saying "life deserves care, hence if we touch it (through synthesis), we do so with responsibility".

When the three theological assumptions fall, transcendent creator removed by immanence, life as gift removed by emergent material ontology, sacred as ontological property removed by cultural critique, , the "playing God" objection reveals itself not as a valid metaphysical argument, but as linguistic anxiety. The phrase remains, possessing emotional power, cultural resonance, , but its anchoring in truth has been removed.

This means not that anxiety is irrational or ought to be discarded. Anxiety regarding new powers is rational. The capacity to synthesise life is genuine power, and all new power deserves serious deliberation regarding its consequences. But anxiety is no argument. It proves not that synthesis is a metaphysical transgression. It merely expresses discomfort before a capacity seeming to exceed limits the human traditionally respected.

The crucial transformation is this: the objection ceases to be theological and becomes technical. The question is not "are we permitted by transcendental law?". It is "is it prudent? Is it responsible? What are the risks? How do we manage them?". These are questions rational deliberation can address; questions ethics, politics, and technique can answer. In dissolving the theological objection, space opens for genuine deliberation.

1.3. Technique as Reflexive Nature

If the theological objection to the synthesis of life does not hold, if there is no transcendent creator monopolising creation, if life is no received gift but an emergent material regime, if sacred is no ontological property but a cultural construction, , how are we to think responsibly the technical creation of life? The answer resides in repositioning the relation between technique and nature: not as opposition, but as nature become reflexive.

Modernity inherited from classical tradition a dichotomy: physis (nature) versus techne (technique). Aristotle viewed nature as that whose principle of movement is within itself, the seed "naturally" grows into a tree. He viewed technique as that whose principle comes from outside, the builder "technically" constructs the house because the principle (the plan) is in the mind of the builder, not in the stone.

Modernity radicalised this dichotomy into an apparently absolute opposition: natural versus artificial. Natural is that which exists independently of human intervention, obeying merely laws of nature, which would have existed even had the human never appeared. Artificial is that which is manufactured, depending upon human will and intention, which would not exist without human intervention. The dichotomy seemed sharp and abyssal. Nature was the realm of determinism, necessity, blind mechanism. Technique was the realm of intention, design, purpose.

Yet this dichotomy never functioned in practice. Two philosophers, Simondon and Latour, dismantle it appropriately.

Gilbert Simondon (1924–1989), a neglected French philosopher, offers a radical critique of the natural/artificial dichotomy. For Simondon, technical objects are no denatured creatures, no exceptions to natural order. They are nature reorganising itself according to determined principles. The machine is no artifice violating nature; it is nature become reflexive, developing the capacity to organise itself according to intention.

Decisive example: water turbine versus natural waterfall. A waterfall is a natural phenomenon, water under gravity descending a slope gaining movement. A turbine is a machine designed to explore this same movement and convert it into energy. What is the ontological difference? None. Both obey the same laws of physics. The waterfall is a non-intentional geological process. The turbine is a process resulting from human intention. But at the substance level, the laws governing movement and energy, there is no difference. Both are nature.

Application to life synthesis is direct: natural cell versus synthetic cell. Natural cell, E. coli, organises according to non-intentional natural selection pressure. Synthetic cell, Syn3.0, organises according to human intentional design. What is the ontological difference? Difference of genesis (evolutionary process versus rational design), not of ontological status. Both are physico-chemical systems metabolising, replicating, varying. Both obey the same biochemical laws.

Technique is not separate from nature. Technique is nature prolonged: nature having developed the capacity for reflexivity, capacity to represent itself to itself and reorganise according to that representation. The machine is the result of this development. The synthesis of life is an extreme extension of this development, the moment when nature (through its human mode) becomes capable of redesigning itself in the biological domain.

Bruno Latour (We Have Never Been Modern) offers a complementary argument. The modern separation between "pure natural" and "pure artificial" was never real, it is a fiction modernity created to distinguish itself from tradition. Yet in fact, there was always constant hybridisation.

The ozone hole, natural or artificial? It is an atmospheric phenomenon (natural) caused by human CFC emissions (artificial): a laboratory construct becoming property of a planetary system. The AIDS virus, natural or cultural? An organism (natural) whose transmission is governed by sexual and blood practices (cultural): biology interwoven with history and social practices. Cultivated maise, natural or artificial? An organism (natural) subjected to ten thousand years of human selection (artificial): a hybrid that would not exist without human intervention, yet completely alive and natural in its biological functioning.

Latour maintains: modernity insisted upon a purity that never existed. Everything modernity called "pure natural" (climate, wild biology, ecosystems) was always already affected by the human. And everything modernity called "pure artificial" (machines, cities, institutions) participates in natural order, obeying laws of physics, ageing, death.

The synthesis of life is no creation of something genuinely "artificial" violating "pure nature". It is the extension of a hybridisation process that always occurred. When we synthesise a cell, we do explicitly what we always did implicitly, marking human intention upon biological processes. The innovation is merely that marking is now direct and controlled, not oblique and unconscious.

Peter Sloterdijk (You Must Change Your Life) radicalises further: technique is not adventitious to the human; technique is constitutive of the human. There is no "natural" human prior to technique. Language is technique (exteriorisation of thought). Education is technique (character formation). Ascetic practices are techniques (body discipline). Medicine is technique (bodily care). Domestication is technique (nature control).

This means: the human is no animal that subsequently developed tools. The human is an animal always-already technically manufactured. We are creatures of technique, this is our biological specificity. Capacity for language, planning, cultural transmission, all this is technique. The human we imagine as "natural" (pre-technical) is a retrospective fiction.

If the human is a technical creature, then the synthesis of life, an intensely technical operation, is no violation of human nature; it is the realisation of human nature. Synthesis technique recruits capacities specifically human (rational design, intelligent manipulation, long-term planning) and extends them to the biological domain. This is no transgression; it is a natural extension (in the sense of "expected" and "coherent with what the human is") of our technical capacities.

When the natural/artificial dichotomy is dissolved, when technique is recognised not as exterior to nature but as nature become reflexive, , the synthesis of life ceases to appear as a boundary violation. It appears as the realisation of a capacity nature (through its specifically human mode) always contained: the capacity to comprehend and reorganise itself.

Creating life in a laboratory is not defeating nature; not imposing human will against natural order. It is nature, having developed reflexivity and intention in us, reorganising matter in a new mode. The laboratory is no world apart, separate from nature; it is an ecosystem where nature continues operating (bacteria, enzymes, chemical energy), now under intentional direction.

This means not that synthesis is without constraints or responsibility. It means constraints and responsibilities are not theological (we violated no sacred law). They are pragmatic, ethical, ecological. Synthesis functions or fails according to criteria nature itself establishes: cell metabolises or not, replicates or not, integrates or not into an ecosystem. Responsibility is the care an agent holds for what it creates, not because violating a transcendental interdict, but because the agent belongs to the fabric it transforms.

1.4. The Legitimate Ethical Objection: Responsibility, Not Transgression

The dissolution of the theological objection means not that the synthesis of life is problem-free or without ethical constraints. On the contrary: when the theological barrier falls, a genuine ethical objection emerges, of a completely different nature. It ceases to be "do not create because you violate sacred law". It becomes "if you create, you are responsible for the consequences".

This transformation is crucial. The theological objection offered comfort of a particular type: if the human simply abstained from synthesis, responsibility would be satisfied, abstention satisfied transcendental law. Responsibility was negative: doing nothing. The ethical objection, conversely, radicalises responsibility: no abstention satisfies automatically the demand; merely continuous vigilance over action consequences.

Hans Jonas (The Imperative of Responsibility, 1979) diagnoses a decisive situation: modern technology created for the first time in history a situation where acting irresponsibly is ontologically dangerous, not merely ethically bad, but a threat to the material conditions of human and biological life. Pre-modern technique was reversible: if you built a wrong house, you could demolish it; if you planted a crop failing, you could sow again. Impact was local, temporal, repairable.

Modern technique, nuclear, genetic, climatic, is irreversible and planetary. You can construct weapons exterminating civilisation; alter atmospheric composition drying continents; synthesise organisms escaping and colonising ecosystems. Effects exceed the agent's temporal horizon (affecting future generations), spatial horizon (affecting the whole planet), and epistemic horizon (unknowable total consequences). Consequence: responsibility ceases to be an ethical option; becomes a condition of survival.

Jonas formulates the responsibility imperative: "Act so that the effects of your action are compatible with the permanence of genuine human life on Earth." Says not "do not use technique"; says "when using technique at a scale altering life conditions, do so with continuous concern for long-term consequences".

Synthetic biology is modern technique in Jonas's sense. You can synthesise organisms spreading uncontrollably; create bacteria competing with native species; open bioterrorism possibilities if genetic synthesis becomes accessible to unregulated groups. Consequence: responsibility mandatory, no optional virtue; condition of any responsible synthesis.

Jonas extends: every living organism is metabolism. No accessory functionality; proper mode of the living, continuous exchange with environment to maintain structure against disorganisation tendency. Human no exception; metabolises too. But human metabolism amplified by technique, extracting resources, transforming, discarding on planetary scale. Responsibility born here: part of planetary metabolism continued; organism within organisms; transforming fabric made of.

Life synthesis metabolic operation: reorganising living matter per intention and calculation; belongs to same planetary metabolism suffered. Hence: responsibility is recognising integration; no exterior observer; process acting inside systems lived, inhabited, depended upon. Consequence simple: any synthesis harming ecosystem harms conditions of own living.

Shelley offers lesson Jonas formalises centuries later: crime not creation; crime abandonment, refusal of responsibility for created. Victor punished not for synthesising creature; punished for denying care. Creature sensitive, intelligent, desiring recognition; tragedy results entirely from rejection, creator denying created community, education, recognition.

Had Victor acted otherwise, recognised creature, offered educational care, integrated into community (even of two), accepted continuous responsibility, , story different. Creature suffered less; possibility of meaningful life. Transforms narrative meaning: no creation condemnation; condemnation of creation without ontological commitment to created.

Post-theological responsibility not abstention; active, continuous vigilance: epistemological vigilance (what know of created effects? what ignore? what research necessary to reduce uncertainty?); practical vigilance (what constraints impose on synthesis to contain risk? what international regulations necessary?); ethical vigilance (what care offer created beings? what rights recognise?).

Vigilance never ends: no point stating "responsibility fulfilled, can relax". World changing; knowledge incomplete; new risks emerging. Responsibility continuous process of learning, adjustment, regulation.

Three responsibility domains emerge:

  1. Toward the creature: If synthesis produces integrated living being, being possesses capacity to suffer (if strong reasons to believe). Hence: responsibility not to inflict unjustified suffering. Means: create not organism capable of conscious experience unless strong reason believing well-being, not merely existence. Open question, boundary between non-suffering chemistry and suffering biology unknown.
  2. Toward the ecosystem: Synthetic being enters ecosystem. Can compete with native species? Spread uncontrollably? Alter food web dynamics? Responsibility to contain risk via control mechanisms (death genes, rare nutrient dependence).
  3. Toward knowledge: Synthesis reveals things about living previously ignored: unknown function genes, regulation mechanisms uncomprehended, redesign possibilities unsuspected. Responsibility to integrate new knowledge into global living comprehension, avoid harmful use of new knowledge.

Theological objection dissolved; question reposed in new register: "is it permitted?" (theological) becomes "is it responsible?" (ethical). To answer, comprehend: can we effectively create life? What knowledge synthesis offers? How operate in practice?

Section 2 addresses via concrete cases, Venter's minimal genome, Elowitz-Leibler synthetic circuits, Arnold's directed evolution: cases showing synthesis not theoretical speculation; functioning knowledge mode: constructing is comprehending. Synthesising life confronts questions pure theory never posed, answered empirically.

2.1. Epistemological Genealogy: Verum Ipsum Factum

The idea that constructing is knowing, that making institutes truth, emerges not from contemporary synthetic biology as its invention. Possesses genealogy tracing back centuries, requiring full recovery because without it one comprehends not why life synthesis constitutes genuine investigation generating new knowledge rather than applying prior knowledge to a new domain. Synthetic biology rests on this Vichian principle, verum ipsum factum, , mapping this thought tradition with precision.

Giambattista Vico, eighteenth-century Neapolitan philosopher, inverted radically the epistemological order René Descartes established as foundational for intellectual modernity. Descartes rested modern epistemology on an intellectualist foundation: secure knowledge emerges from clear and distinct ideas in the mind, rational evidence imposing itself by net clarity immanent to representation. Cartesian method promised absolute certainty because intellectual clarity cannot deceive, clear idea corresponds to real or is first-order reality, truth unneeding mediation. Vico contested program from root, not via direct logical refutation, but radical repositioning of truth criterion: true is not thought clearly, , what made, constructed, instituted materially. Verum ipsum factum, the true is the made itself. Brief formulation, first-order epistemological consequence: genuine knowledge belongs to that whose genesis controlled integrally, produced from foundations where no element escapes because instituted materially.

Vico offers two-part argument: First, God knows physical nature perfectly because created it, God total creator, whose will established every cosmic configuration aspect; divine knowledge access integral genesis participation. Second: We, not creating fundamental universe matter or physical laws, possess indirect nature knowledge: observe effects, construct explanatory hypotheses, deduce general laws via inductive procedure. Observational mediation never offers certainty type creative participation confers. Nature knowledge fundamentally approximative, progressively refined via observation accumulation, never complete as participatory comprehension characterising divine knowledge. Yet, crucial step Vico formulated clearly, knowledge of what we construct ourselves is absolutely certain because participating integrally in production from absolute beginning. Mathematics true knowledge because we invented axioms, definitions; Euclidean geometry true because we established point, line, plane definitions and developed consequences; civil institutions and political structures comprehensible totally because built via convention and aggregated action. Knowledge and making coincide absolutely: knowable in totality is capacity to make, genesis responsibility integral.

Implication Vico developed not integrally, latently remaining in program as historical realisation awaiting possibility, : if extending "making" to domains previously appearing ontologically forbidden or inaccessible, synthesising what nature offered as factual data passively accepted, , gain knowledge access Vico reserved to human intellectual or institutional creations. Transform approximative, observation-mediated knowledge into participatory, constructive knowledge. Artificial life synthesis would be late fulfilment of Vichian program promise: extending to living material nature absolute intelligibility previously possessed only over mathematical, architectural, institutional creations.

Epistemological turning point connected to Gilbert Simondon's "technicity". For Simondon, technique is not applying theoretical knowledge to inert matter; technique is a genuine existence mode, mode through which material reality transforms. Technical act not separable from embodied knowledge, is knowledge form. Synthesising genome: not "applying biochemistry knowledge"; embodying knowledge in practice, converting theoretical intelligibility into material operation. Craig Venter synthesises whole genomes: not merely applying; inaugurating new knowledge mode where theory and practice coincide.

Francis Bacon, centuries prior to Vico, situated knowledge not in abstract mental representation, but power, capacity to act and transform. Scientia potentia est, knowledge is power, capacity to act, modify, control. Genuine knowledge manifests not in passive contemplation of natural phenomena; manifests in capacity to control, modify, manipulate, exercise selective domain over material conditions. Scientific experiment not passive observation recording; active perturbation, interrogation via experimental violence, forcing nature to reveal secrets via deliberate interventions, imposed conditions, manipulated contexts. Baconian investigator sits not peacefully observing; forces nature to answer via artificial condition construction, controlled experimental frameworks, coercing properties unmanifested in unperturbed state. Laboratory: space where nature interrogated violently, intelligently to speak truth unoffered voluntarily.

Richard Feynman summarised Baconian program in canonical formulation: "What I cannot create, I do not understand." Simple appearance until thought with philosophical rigour. Feynman proposed not knowledge subjective opinion or individual preference; positive, empirically testable statement on genuine comprehension nature. Comprehension manifests, proves in capacity to construct analogues, reproduce phenomena under precise controlled conditions, perturb systems, observe responses predictably matching theoretical predictions. Experimentalism not passive observation; active construction of new unexisting situations, testing if theoretical representations anticipate behaviour of new instituted systems. Reconstructing phenomenon under absolute control, making, , acquire certainty comprehension not speculative fiction matching observation accidentally; practice functions, constructed exhibits behaviour theoretical prediction stated. Feynman genuine comprehension: capturing operative essence via precisely controlled replication capacity.

Contemporary biological engineering retrieves, radicalises Vichian-Baconian-Feynmanian program completely. Craig Venter, conceiving first artificial genome synthesis (Syn1.0 in 2010; Syn3.0 in 2016), applied not "conventional molecular biology knowledge". Converted biology from observational/theoretical science, investigating existing organisms, to constructive science, creating new unexisting organisms. Synthesis no logical outcome derived from complete theory; original investigation results surprising in construction, revealing deep knowledge gaps no accumulated observation of pre-existing natural organisms rendered visible with destructive precision. Synthesising autonomous living cell from raw chemical components, nucleotides, proteins, lipids, , answer question theoretical biology never formulated with sufficient rigour for experimental answer: what components necessary, sufficient? What genetic structures indispensable for cellular life? What functions suppressed, removed, eliminated without cell collapse? Construction forces living matter to reveal fundamental ontological distinctions via behaviour, organism lives or dies, functions or fails, unmediated by interpretation.

No incremental refinement of established scientific methodology; first-order epistemological revolution redefining knowable, knowledge institution, scientific investigation nature. Vico correct: true is made. Synthesising life, whole genomes placing nitrogenous bases in pre-defined sequence, constructing new biological circuits unexisting anywhere until synthesis moment, modifying organisms systematically, , gain intelligibility access otherwise permanently inaccessible, barred to observation. Power to know what previously described, theorised at distance, external observers condemned to approximation.

Knowledge biological engineering offers not application of prior theory to practice, , generation of new, unforeseen knowledge via constructive act, revealing real structure instituted by making as true knowledge.

2.2. The Minimal Genome: Constructing to Discover

When Craig Venter, Daniel Gibson, and the JCVI team synthesised Syn3.0 in 2016 (cellular life "minimal genome"), primary objective not "replicating" living organism via chemical synthesis. Converted ancestral theoretical biology question, what essential to life? minimal sustainable material essence?, into empirically investigable, resolvable question via systematic iterative construction, continuous testing. Operative question: reduced gene set supporting living, autonomous, functionally integrated cell, capable of continuous metabolism, genetic replication, cell division?

Answer revealed via iterative chemical synthesis, cell viability testing, results analysis, dispensable gene removal: 473 genes. Context: E. coli contains $\sim 4,600$ genes; Saccharomyces cerevisiae $\sim 6,000$ genes; human $\sim 20,000\text{--}25,000$ genes. Syn3.0 cellular life reduced to $<10\%$ model organism baseline genetic complexity. Yet cell functions integrally: metabolises glucose via complex enzyme cascades; replicates DNA with fidelity; divides cytoplasmically into two viable daughter cells; reproduces generationally. No functional collapse; no obvious biological incompleteness; dies not of ontological deprivation. Alive by definition mattering ontologically, operatively: executes continuous metabolic cycle, maintains physico-chemical homeostasis, reproduces with faithful genetic information transmission.

Perturbing revelation: construction left open permanent comprehension gap, $\sim 149$ genes in Syn3.0 ($\sim 31\%$ total genome) possess completely unknown biochemical, biophysical, operative function. No one currently explains with complete mechanistic detail why each gene necessary for cellular survival. Cell survives not without them, experimental fact: remove any of 149 genes, cell viability collapses; cell dies. Specific molecular function, biochemical operative role, action mechanism remains opaque, inaccessible to contemporary chemical, biophysical analysis. Revelation Vico predicted, program promised: construction permitted making life, revealed impoethed precision contemporary theoretical comprehension limits, exact points where knowledge ends, deep ignorance begins.

Method making discovery possible radically different from historical observational biology. Traditional observational approach: investigator studies sequenced natural genomes, sequences DNA, seeks analogies with known function genes, catalogues genes systematically, correlates genotype with observed phenotype, infers function via homology with characterised genes. Methodologically powerful, responsible for modern biochemistry, twentieth-century molecular biology rise. Constitutive inamovible limitation: natural organisms, product of billion-year evolution under selection pressure, carry genetic "dead weight", historical redundancies, functional duplications uneliminated because non-detrimental to reproductive survival. Genes indispensable in ancestral lineage dispensable in contemporary population via alternative pathway evolution, remaining in genome because natural selection lacked opportunity, selection pressure to remove without unacceptable cost. Evolved organisms never spartan in genetic efficiency, carry evolutionary baggage, vestigial genes, historical inertia elements.

Synthesis breaks evolutionary inertia completely, radically. Begins from absolute zero, fundamental chemical molecules, technical capacity to synthesise in pre-defined sequence, and removes systematically, iteratively, dispensable survival elements. Investigator asks not "what function has this specific gene?", theoretical question directed to genomic databases, literature, evolutionary conservation assumptions. Asks directly, operatively: "can cell live without this gene?", practical, factual question directed to living cell matter answering with viability or death, unadmitting mediated interpretation. First question speculative, offering multiple answers depending on theoretical assumptions, available data interpretation. Second question binary, factual, merciless, inescapable: lives or dies. Factual answer depends not on theoretical consensus, nomenclatural convenience, philosophical preferences on life definition/essence; answer given by cell itself, objective material behaviour: lives or dies.

Iterative synthesis method revealed genes uncomprehended in isolation. Venter discovered genes dispensable tested isolated, essential combined with variations in other genes. Reveals relational network complexity: genes operate not in absolute isolation, but relational contexts where redundancies, alternative pathways, compensations matter critically. Construction reveals reduced essence (minimum gene number), relational interdependence structure, cannot comprehend gene truly without complete relational context inside total functional system.

Inverts knowledge/ignorance relation in contemporary scientific context. Traditional ignorance vague, invisible, dispersed: "aspects of cellular biology uncomprehended", "mechanisms remaining unknown", "processes where knowledge incomplete". Formulations locating no precise gap, indicating no clear future investigation direction, leaving ignorance general field quality without structure. Ignorance revealed by biological engineering precise, circumscribed, navigable, territorialisable: "know genes X, Y, Z and 146 others ($\sim 149$ specific, complete list) necessary for autonomous cellular life, but unknown specific biochemical function, molecular operating mechanism, homeostasis/cellular organisation role". Ignorance no failure/deficiency; second-type knowledge, knowledge about unpossessed knowledge, gap structure.

Craig Venter synthesised Syn3.0 not because biological knowledge attained completeness where technical application of established results remained; synthesised suspecting theoretical comprehension radically incomplete precisely in dimensions construction rendered visible. Genuine Vichian making nature: demonstrates construction capacity, offers technical possibility confirmation, reveals precise intelligibility limits construction leaves open, offers clear map for subsequent investigation. Synthesis functioned; cell lives; cell remains theoretically opaque in 31% of genome. Success revealing simultaneous failure, success creating life, failure completely comprehending created. Fruitful tension where true investigation thrives.

2.3. Synthetic Circuits: Testing Models Through Construction

Molecular biology developed theoretical, mathematically articulated genetic regulation models describing how DNA molecules bind specific proteins (transcription factors), how molecular bindings affect transcription speed, how proteins bind in signalling cascades. Mathematical sophistication considerable, ordinary differential equations describe temporal dynamics, Boolean networks encode binding logic, stability analysis predicts long-term behaviour. Methodological problem no theoretical refinement resolves: in living natural organisms, each gene immersed in untangleable network of thousands, millions, of molecular influences: epigenetic factors modify DNA accessibility; molecular-level stochastic noise introduces unpredictable random fluctuations; non-specific protein interactions create parasitic bindings; historical evolutionary pressures left residual genome marks unpermitting discrimination between necessary and conserved by inertia. How verify epistemological rigour if theoretical model captures mechanistic essence described, or converges accidentally to observed nature behaviour via multiple error compensation cancelling out?

In 2000, Michael Elowitz and Stanislas Leibler proposed an elegant, ontologically perturbing solution: construct a synthetic genetic circuit as described by the theoretical model in pure form, without complications from evolutionary complexity accumulated over millions of years. Result called repressilator, synthetic circuit of three genes mutually repressing in closed loop configuration: Gene A codes repressor protein blocking Gene B transcription; B represses C; C represses A. Loop closed: A represses B, B represses C, C represses A. What succeeds when instituting structure? Theoretical model, based on molecular kinetics equations and genetic expression, predicts spontaneous periodic oscillation emerging from system. When A active (transcribed, translated), protein accumulates, B silenced. As A protein decays via normal degradation, B no longer repressed, starts transcription. As B protein accumulates, C repressed, falls. As C falls, A no longer repressed, rises again. Cycle completes, restarts. Mathematical modelling result: gene expression oscillates periodically, period and amplitude determined by repressor proteins kinetics, degradation rates.

Constructed experimentally in laboratory: integrated synthetic repressilator genetic sequence into E. coli genome; coupled green fluorescent protein (GFP) to one circuit gene, glowing green when gene active, transcribed. Observed under microscope: bacterium blinks, turns on green fluorescence with predictable period, turns off, turns on again in quasi-periodic rhythm. Dramatic confirmation theoretical model captured genuine underlying molecular physics regarding genetic regulation. Theory no speculative fiction, corresponds directly to real molecular operation structure.

Deeper revelation emerged measuring behaviour with quantitative rigour: oscillations significantly noisier, irregular, than pure deterministic model predicted. Substantial stochastic variation, random timing, amplitude, phase fluctuations systematically deviating from theoretical predictions. Exact reason: circuit operates with very small molecule numbers. Each repressor protein exists in reduced quantities, dozens, hundreds of copies per cell, not thousands or millions. Small molecule numbers: random effects, quantum binding/unbinding rates fluctuations, protein degradation time variation, Poisson distribution molecular birth/death events, become visible, substantial at whole-cell scale. Natural E. coli observation never revealed this clearly, isolated: mixed inextricably with thousand other cellular noise sources, other genes expressed, operating circuits, varying osmotic pressure. Minimal synthetic circuit: stochastic noise emerges pure, isolated, quantifiable, impossible to ignore.

Model confirmed qualitatively, oscillations exist, , disconfirmed quantitatively in magnitude aspects. Invalidates not theory; refines critically. Revealed theoretical model, capturing correct qualitative logic, incomplete in fundamental assumptions: implicitly assumed large molecule regime where stochastic effects neglected as approximation error. Cell numbers small, stochastic effects significant, modifying observable behaviour. Deliberate synthesis of new circuit permits investigating regime non-existing naturally with this purity, isolation degree.

Repressilator initial application example. Synthetic genetic circuit synthesis revealed biological organisation principles natural genome observation never isolated clearly. Ron Weiss built "biological digital logic", circuits computing Boolean operations (AND, OR, NOT) via simple genetic regulation. Circuits "decide" biochemically, taking molecular input decisions, generating specified functional outputs. Reveals for life comprehension: computation, propositional logic, algorithm information processing capacity not exclusive system nervous/mental consciousness privilege, fundamental material operation biological molecules execute organised appropriately under specific constraints. Deliberate synthetic circuit construction demonstrates possibility, , forces radical reinterpretation of cognition, "intelligence" meaning in pre-symbolic living matter context. If regulator gene sequence executes Boolean logic, computation and information processing capacities distributed across material networks, unconcentrated in unit conscious organ. Natural organisms observation, where biological logic operates coupled to thousand simultaneous processes, never revealed generalisability with isolation sufficient for true comprehension.

CRISPR gene editing, synthetic uses equally revealing. Jennifer Doudna, Emmanuelle Charpentier recognised bacterial immune mechanism (phage defence bacteria evolved) reconstructible ex vivo, purified, applied as single-nucleotide precision gene editing tool, , discovered not merely technique; demonstrated genetic material appearing fixed, ontologically immutable in natural complex organisms, synthetically manipulated ontologically plastic. Rewriting genetic sequences with page text rewriting ease forces fundamental reinterpretation of "nature" meaning. Genome plasticity, essential malleability, truth natural living organism conceals via integrated functioning inertia, complexity. Synthesis reveals fundamental, immanent property.

Fundamental lesson. Drew Endy, synthetic biology establishing lead investigator, argues creates new science with own epistemic criteria, irreducible to observation, theorisation isolated. Synthetic circuit not "better" than natural living organisms observation, error thinking hierarchical competition; epistemically, ontologically different. Reveals things observation leaves in shadow by method. Synthesis isolates variables careful observation uncouples not from natural complexity. Synthetically remove lateral influences, auxiliary circuits, evolutionary redundancy, maintain desired core. Observation condemned to study existing, inside nature established constraints. Synthesis creates scenarios nature never explored (lacking selective pressure). Together, newly designed system experimental construction, evolved natural organisms observation, systematic model theorisation, form triangulation offering un-isolated availability intelligibility. Construction theory test as rigorous as observation, complementary.

Making and observing not conflicting epistemological opposites, , complementary reality access modes, each revealing structure, functioning aspects other leaves hidden. Vico intuited, contemporaneity confirms in practice.

2.4. Ontology Through Construction

What knowledge type biological engineering offers practising investigator, observing world? Not merely technical, instrumental knowledge, means, practical knowledge on producing desired effects, manipulating material components for independent predefined goals. Mere prior knowledge application to new context. Primary first-order ontological knowledge: genuine discovery of what a thing is in fundamental material structure, true constitution, indispensable identity maintenance properties versus dispensable, contingent properties, functional dimensions articulation forming operative integration.

Ontology transformation into experimental discipline far-reaching philosophical revolution. Centuries "what is X?" question condemned to speculation, debated in seminars, texts, logical arguments never attaining certain conclusion, , engineering offers new method: question formulated operatively, answered materially. Makes ontology not trivial; precise, constraining, responsible. Synthetically making life, hiding behind vague definitions impossible: created cell speaks via behaviour whether "life" understanding adequate, incomplete. Transformation in "comprehending" meaning.

Canonical ontology question spanning philosophy history: what essential to X? What properties X loses not without ceasing to be X? Constitutive true essence versus accidents, contingent properties? Life domain: what essential to life? What life ceases not to be without ceasing to be life? Constitutive life concept properties? Traditionally answered via rigorous conceptual analysis, definitions proposed: "life is what metabolises", "auto-reproduces", "responds to stimuli", "maintains internal complexity against universe entropic tendency". Arguments over boundaries, category borders. Answer remains language convention, discursively constructed agreement. Residual indetermination conceptual analysis resolves not, question whether definition captures true essence or prominent accidents appearing essential.

Biological engineering offers different ontological investigation method: experimental, constructive, factually testable. Essential to life operatively: what unremovable from system without immediate organism death. Operative definition empirically testable, precise, irrefusable; depends not on arbitrary conceptual stipulation. Craig Venter removed systematically Syn3.0 genes, cell answered directly: lives or dies. Unignorable, reinterpretable, relativisable, contestable answer. First-order material data resisting theoretical preference. Cell offers no "opinion" on gene essentiality. Cell viability material fact imposing independently of philosophical preferences, conceptual definitions.

Engineering converts speculative question into empirically resolvable question via systematic construction. Resolving experimentally, discovers life essence not simple, unidimensional as prior theories suggested: not "merely metabolism", "reproduction", "variation capacity". Multidimensional integrated complex where components articulate necessarily: continuous chemical flux maintaining energy gradients, electron transfer (operative metabolism); precise, non-perfect genetic code replication (controlled variation reproduction); programmed cell death (apoptosis) maintaining population renewal; differential cell specialisation (multicellular) executing different subroutines. Remove fundamental components, system collapses in death. Genuine ontological discovery: life essential structure restricted, specific, physically determined in constraints than prior theories/analyses supposed/revealed.

Engineering reveals uncomprehended, offers surgical ignorance localisation. 149 unknown biochemical function genes in Syn3.0 tangible testimony ontological knowledge fundamentally incomplete post-successful synthesis demonstrating life possible without known function components. Vague formulation "things about cellular biology uncomprehended" gives false comfort without instructing/locating gap. Precise, territorialised statement: "know X, Y, Z and 146 genes ($\sim 149$ specific, complete list) necessary for autonomous cellular life, but unknown specific biochemical function, molecular operating mechanism, homeostasis/organisation role". Ignorance no engineering failure, , different ontological success: installing permanent responsibility. Learned ignorance location with surgical precision: material functionalities, biochemical operations unnamable, undescribable, unexplainable, life absolutely depending upon for functioning.

Localised, precise ignorance revelation crucial for contemporary scientific responsibility. Insufficient saying "comprehended because constructed and functions". Say: "constructed and functioned, but functioning revealed comprehension incomplete than anticipated, responsible recognising incompleteness honestly, pursuing investigation with renewed rigour". Ethical responsibility ends not at synthesis act; begins there, successful construction meeting limits, recognising gap. Synthesis question opening, not investigation closure. Synthesising life, attaining functioning, acquire permanent responsibility comprehending un-attained, investigating 149 unknown functions mechanisms, attentive to unpredicted effects, maintaining vigilance over created.

Final consequence: ontology, millennium speculative discipline, ceases exclusively discursive domain. Fundamental ontological questions, "essential to life?", "reduced minimal dispensable structures?", "properties varying/suppressible without collapse?", empirically investigable, resolvable via systematic construction. Synthesis offers essence access method unavailable to isolated conceptual investigation. Resolves not all being questions, leaves open, 149 unknown genes function demonstrates. Offers different rigour, non-purely logical: matter responding to constructs, confirming/refuting expectations, imposing precise constraints over possible, necessary, contingent. Life ceases untouchable mystery; becomes open question construction permits exploring newly, precisely, responsibly.

Synthesis biological not new methodological instrument available to investigator, revelation that genuine living nature comprehension required constructive act. Isolated theory, accumulated observation, mathematical modelling, conceptual analysis, condemned by structure to leave gaps. Theory operates at observed, categorised, named level; remains reactive. Synthesis proactive, brings being unexisted, forces reality to answer unexisted questions. Permits questions theory formulated not (no examples to interrogate).

Craig Venter succeeded Syn3.0 living, 473 genes, 149 unknown function, , applied not existing molecular biology theory. Created new experimental situation: biological reality forced to reveal truths nature observation never revealed. Synthetic cell functions: synthesis possible, theoretical knowledge existed permitting. Cell remains mystery in 31% of genome: theory incomplete, construction revealed incompleteness no observational analysis achieved. Genuine knowledge, non-prior assumption confirmation, localised, precise ignorance discovery.

The truth is the made, and the made revealed truth isolated theory never attained. Biological engineering institutes new ontological knowledge mode, where real comprehension measured by capacity to make, investigation participation in real material genesis, responsibility permanent comprehension limits recognition.

3.1. The Creator Belongs to the Category of the Creature

We pass now to a completely distinct territory. We no longer question the legitimacy or possibility of synthesis, resolved: legitimate, epistemologically viable, genuine knowledge construction tested in matter. Question moving section ontological, radical: who is the we making synthesis? Synthesising agent status? Creator nature creating? Question, posed seriously, repositioning living creation comprehension.

Spinoza offers starting point dissolving irreversibly creator/creation separation. Deus sive Natura, God is nature, no transcendent entity governing outside. No separate domain, no creator ontologically separate from created. Continuous immanence, uninterrupted flux where everything material process determining by own nature. Matter ungoverned by external will, non-passive substance shaped by transcendent intelligence. Reorganises under constraints faced, physical, chemical, biological, environmental pressures. All inside unique, radical immanence. Spinoza refuses Cartesian dualism (mind separate from matter), theological transcendence (God outside nature). Single universe, substance, immanence plane where everything occurs.

Human, Spinoza topography: particular immanence mode. Process reorganising matter forms other processes reorganise not, incorporated intelligence, material intention, neural network anticipating/planning capacity. Extraordinary complex feedback system, memory spanning centuries via writing/cultural transmission, consciousness reflecting upon self. Completely immanence part, non-outside nature in fundamental aspect. Nature attaining reflexivity point, matter operating reflexively upon self, comprehending, modifying.

Complete in Spinoza: no separation possible creator/created (both modes, particular expressions, of same substance). No human exception, no ontological privilege placing outside matter. Creator difference: mode reorganising other modes, process affecting other processes, , remaining mode, integral immanence part.

Applying to life synthesis radically changes creation comprehension. Human immanent, non-outside nature, non-transcendent agents, , synthesis completely immanent operation. No immanence violation from outside; no transcendent law transgression prior to action. Transformation occurring inside unique immanence, material process continuing, prolonging, amplifying other material processes. Life synthesis gesture living executes upon self, reflexively, consciously, intentionally, life folding upon self to reorganise. Life creating life, inside unique immanence, without creator/created ontological separation. Creator belongs completely to creature category. Both same matter modes in continuous transformation, life expressions continuing eternal reorganisation.

Radical reconfiguration of technique, creating meaning. Bergson, Book I: life continuous tendency creating new forms, expanding, opening unexisting possibilities. Technical human no exception, most conscious, reflexive realisation. No human privilege standing outside life operating sovereignly. Radical participation in life nature: continuous transformation, matter reorganisation, unexisting possibilities creation. Technical human extends via physical/cognitive tools, symbolic language permeating action, writing memory spanning generations. Fundamental logic same as biological process: matter reorganising per intelligence applied to faced constraints, found resources, experienced pressures.

Simondon offers precision philosophical instrument completing Spinoza: technique not separate from nature, categorical statement placing correctly. Technique no artificial domain violating pure/pristine natural domain before technical intervention. Machines, technical artefacts, constructed organisms no denatured creatures corrupting original nature. Nature reorganising self via engineering, physical/chemical processes intentionally selected, oriented, potentiated by human intelligence. Machine product of material operations directed by planning, designing, imagining, testing capacity. "Directed by intention" renders machine no less natural, renders exemplar of how nature operates passing through intelligence. Machine reveals matter containing directed transformation capacities, improvement possibilities per constraints/goals, latent potentiality awaiting intelligence articulation.

Synthetic life inherits Simondonian logic completely. Not "natural life corrupted by technique, losing original purity". Life reorganised via technical capacities human developed over millions of years of evolution, tools, planning, knowledge transmission, environment modification capacities. Human is nature developing extraordinary capacity modelling another nature, molecular biology, genetic processes, cellular metabolism mechanisms. Renders synthesis no less natural; renders exploration of virtual possibilities in chemical/biological relations, possibilities inscribed in matter as potential awaiting update. Synthesis places possibilities in actuality; realises capacity present in latency: living mode (technical human) modelling another living mode, creating genuinely new life forms, unexisted forms, amplifying living possibilities domain.

Clear continuity between natural evolution and design, obscure point crucial for synthesis comprehension. Natural evolution selection of random origin variations: unoriented, unintentional, unplanned mutations, persisting when facilitating organism survival/reproduction. Design selection of intentional variations: humans choosing characteristics to preserve, amplify, projecting specific mutations, cultivating lineages per anticipated, comprehended criteria. Operative mechanism fundamentally same: variation persisting when functioning, integrating viability/operatively, rendering organism capable of surviving, reproducing. Difference merely selecting agency: evolution natural selection, environment pressure, competition, differential reproduction of fittest; design human intention, rational comprehension, consequences anticipation, reflexive planning. Both material operations inside chemical/biological constraints; both select immanently; both work with existing living matter. Design evolution continuation, non-rupture/negation. New evolutionary operation form, directed, intentional, reflexive, yet genuine evolutionary operation, selection occurring in common immanence fabric.

Jonas contributes vital dimension un-neglectable in synthesis comprehension. Every living being metabolism, central, structuring concept for adequate, deep life comprehension. Metabolism continuous exchange process with environment, continuous matter transformation maintaining organism organised structure against entropy, universal disaggregation tendency. Accidental process characterising not merely some living beings, proper, constitutive being mode of all living. Organism is what metabolises continuously, transforming matter, converting energy, maintaining organised, differentiated structure. Jonas sees essential for responsibility: metabolism responsibility incorporated in living being itself. Organism responsible for own living, must continue transforming matter, maintain against disorganisation, operate continuously to remain organism, disintegrate not.

Technical human extends metabolism genuinely, non-transcendently. Transcends not biology, amplifies, distributes via material tools, structures continuing matter transformation to maintain life. Life synthesis, metabolist perspective, human metabolism in operation, living organism using metabolic capacities (thought, object manipulation, planning, experimentation) to produce another life, create new living beings. Reorganisation of matter continuing metabolic trend of continuous transformation characterising all living. Synthesis incorporated metabolism mode, non-violation/rupture. Metabolism attaining reflexivity, operating consciously upon own processes, questioning self while metabolising.

If synthesis metabolism, continuous life mode, , responsibility metabolically incorporated: inside being, non-transcendent, non-external, non-imposed from outside. No external law prohibiting/authorising abstractly; possibility condition of technical living. Synthesis causing ecosystem harm, harms own living possibility conditions, affects nutrient cycles, alters matter fluxes depending on ecological balance, compromises biota depended upon continuously. Responsibility metabolically incorporated experience: responsible because living; living means depending continuously upon material transformations involving other living, whole ecosystem as feedback system. No separation possible between action and ecological effects, both occurring in shared metabolic fabric.

The creator is not outside the creature, it is a creature become reflexive, attaining the capacity to operate consciously upon the very bases of its material living.

3.2. Life as Reflection Upon Itself

We enter a deeper, revealing comprehension layer. Creator belongs to creature category, synthesis gesture living executes upon self reflexively, intentionally, , opening radical transformation in life comprehension, living meaning, living being nature. Ontological meaning of living when living capable of reflexively reorganising life processes?

Observation dissolving illusion regarding technique novelty, exclusivity: technique no human invention, historical, conceptual error affirming. Non-exclusive property emerging with human species as exceptionality mark. Bacteria use technique constantly in operations: enzymes nanometre biological machines catalysing specific chemical transformations, unoccurring naturally at same rate, concentration, selectivity, efficiency. Enzymes operate as molecular tools, exponentially increasing reaction speeds, directing chemical processes with extraordinary precision un-attained by random agitation. Pure technique operating at molecular scale. Animals use technique continuously in survival operations: chimpanzees use sticks extracting insects from tree fissures, deliberate appropriation, modification of natural material for specific goal. Birds build ninhos with refined architecture, angle/weight distribution calculation, intuitive resistant structure knowledge. Beavers build dams reconfiguring hydric landscapes, altering water fluxes, creating new habitats, transforming environment radically. Technique everywhere in living, continuous demonstration of life capacity reorganising environment via material instruments, distributed intention in continuous operation.

Human invented not technique, ignorant of natural history affirming. Extended unprecedentedly in life history: via symbolic memory, capacity storing technical knowledge in vocal language, writing, artefacts preserving knowledge beyond individual life; via planning transcending immediate, drawing on paper, abstract thought, future possibilities before material realisation, simulation testing before acting; via intergenerational transmission, teaching another, accumulating innovations, avoiding technical knowledge loss upon individual death; via abstraction, generalisation, comprehending general principles functioning in multiple contexts, applying tested solution to new domain. Spectacular elaboration of capacity life always possessed, , elaboration permitting unprecedented qualification in planet life history, opening radically new possibilities.

Human attained point modelling living entirely synthetically, point no other living mode attained, not because other modes inferior, human developed specific capacities: design DNA sequences, specifying exact genetic base (adenine, thymine, guanine, cytosine) following which, controlling information encoding order; cultivate isolated cells in bioreactors, maintaining temperature, pH, nutrients, oxygen conditions calibrated per cell biochemistry knowledge; synthesise life from molecular bases, combine molecules intentionally, create genuine living organism metabolising, reproducing, responding to environment. Radical reflexivity: reflection transcending theoretical comprehension to become practical self-modification. Life conscious of own operative functions, metabolism, reproduction, genetic variation, cell differentiation, environmental stimuli response, , reconfiguring intentionally; asking precisely: essential to life? dispensable? variations to create? new capacities to introduce? emergent behaviours to explore? Poses questions comprehending via genetic engineering, synthetic biology, research decades, fundamental operating mechanisms.

No living rupture, no life abandonment entering artificial transcendent domain. Living attaining reflexivity degree permitting reflecting upon self, operating upon operation bases without ceasing living. Intelligent matter, life intelligent matter, developing thinking, calculating, anticipating capacities, attaining point examining self with operational precision, deeply comprehending functioning, modifying intentionally without ceasing matter, living, operating materially.

Synthesising life, discover un-discovered with precision, system: life constitutive principles, rendering alive, functioning. Discovered forced to articulate with operational clarity for synthesis functioning, synthesised organism viability. Computational simulation incomplete knowledge, models wrong in unrevealed aspects, reality complex. Synthesis knowledge tested in matter, verified in continuous real-world operation. Design gene unfunctioning as anticipated, learn incomplete comprehension, unconsidered factor. Design biological circuit, mutually regulating genes, recursively influencing, , producing un-anticipated emergent behaviour, discover biology possibilities natural system observation never revealed. Organisms with complexity so great unobserving emergent properties unless synthesising simplified versions, building comprehension bottom-up, components to system. Synthesis forces deep ontological questions observation postpones, ignores. Synthesis superior mode of life knowing self, knowledge incorporated in living matter, tested against continuous operation reality, leaving indelible marks in world.

Haraway offers additional perspective, neglected in life synthesis debates: nature no pure domain prior to culture, illusion obscuring radical reality. No pristine nature state preceding human, human subsequently corrupts via technique. "Nature always already contaminated by culture", deep, inextricable interweaving of natural processes (molecular biochemistry, genetic evolution, natural selection) and cultural processes (millennia agriculture, animal/plant breeding, genetic manipulation, continuous technical modification). Whole known biosphere, living in known landscapes, insoluble imbrication of natural and cultural processes. Wheat selected 10,000 years; cattle bred millennia; wild-appearing organisms products of deep historical human relations: flowers depending on domesticated pollinators, plants germinating exposed to human-ignited fires over millennia. Nature no separate pure domain.

Life synthesis contaminates not "pure nature", continues always-existing contamination, doing so consciously, explicitly, honestly. Synthesised life genuine living being in operation, truly metabolises, reproduces, varies, integrates into ecosystem, competes with organisms. No "less alive" because synthesised, laboratory origin versus wild environment. Alive transparently displaying interweaving with human culture, scientific knowledge, engineering planning. No demerit, deep, radical epistemic honesty: recognising all life always already cultural, "wild" life existing merely in human history ignorance, known life passing through human intelligence, passage beginning millennia ago with plant domestication.

Ethical implication follows immediately, clearly: no pure nature to preserve, "original state" illusion ignoring no original state exists isolated from human, , ethical responsibility consists not in maintaining "original state", restoring "natural purity" un-existing isolated; continuous, vigilant, reflexive care of created mixtures, , recognising living world inhabiting current ecosystem mixture of natural/cultural processes. Life synthesis introduces mixture newly not; continues always-existing mixture, part of all life structure. Doing consciously, intentionally, with responsibility comprehending operating in ecosystem un-isolable from human life/history.

Crucial ontology point changing everything: life synthesis gesture where living, via human technical mode, reorganises living creating possibilities natural evolution never explored, no biological agent created. Exactness: non-"living creating non-living", non-transcendent creator bringing forth radically other, non-living, non-metabolising; "living creating new living", reorganisation remaining inside biological domain, elaboration of virtual possibilities present in living matter genetic/biochemical structure. Synthesis explores possibility space always present, virtual, potential in existing living matter.

Ontological consequence transforming life comprehension: living no domain created in remote evolutionary past, left evolving blindly, un-directed; no kingdom created, left alone for own functioning; domain expanding, opening to new possibilities via multiple continuous operating agents. Natural selection continues operation, environments change, adapting organisms persist; random mutations continue, genetic variation un-directed by intelligence. Now active technical human agency in operation: intention directing variation, exploring genetic possibilities space natural selection never found, natural selection blind, operating merely on chance variation. Synthesis new living agency, non-agency outside life, imported from transcendent/artificial domain; agency life developed, life is, completely immanent to life.

Comprehension poses final question chapter answers not completely, formulates with clarity, exactness for next chapter deep address: if synthesis living gesture, life reorganising self reflexively, , criterion determining if created "really alive"? Property rendering genuinely alive? Origin, prior natural source? Immutable essence, fundamental property separating living from non-living? Reproduction, metabolism, adaptation capacity? Traditional answer for centuries: life origin in prior life, preserving essence across generations, distinct boundary between living/non-living domains.

Synthesis challenges deeply: created clearly alive in operation, genuinely metabolises, truly reproduces, varies, integrates into ecosystem, competes with organisms, , lacking origin in prior life traditionally. Or: origin in prior life, technical human, researchers, engineers, , via entirely artificial mediation, laboratory synthesis, intentional manipulation. Origin non-reliable criterion, synthetic living genuinely alive in operation. Nor essence, synthesis reveals reorganising life components radically different from natural organism, viable organism, operative life, metabolising/reproducing being. Criterion in operation, non-origin/essence. Living defined by what it does, non-what originally/essentially. Question articulating precisely, opening path to next chapter: operative regime defining living?

The living is a regime, not an essence, a continuous operation that can be radically reorganised and still remain a living operation.

3.3. Synthesis as Legitimation and as Radicalisation of Responsibility

Converging now to synthesis of tripartite movement crossing chapter coherently: demolished theological objection blocking synthesis, transcendent creator empty concept, immanence adequate/necessary ontological framework; founded technical knowledge legitimate/genuine mode, engineering non-simple prior knowledge application, new knowledge generation tested in real matter; repositioned responsibility from transcendent moral prohibition to permanent material vigilance, synthesis ontologically legitimate, responsibility non-action abstinence, radical care of continuous action effects.

Life synthesis no transgression. Demonstrated in three non-independent registers forming single argumentative fabric: First, theologically, objection repeating "playing God" rests on removed foundations; transcendent God no real agent operating in universe, no governing will; symbolic fiction losing explanatory, ontological functionality; no transcendent divine will operating in universe synthesis violates (no divine will operating); continuous immanence where all agents, natural, human, technical, operate contiguously, none outside another's domain. Second, epistemologically, engineering legitimate, genuine knowledge mode; non-corruption of "pure" contemplative knowledge, idea of true knowledge from disinterested contemplation, inferior knowledge from interested experimentation ancient ideology to abandon; knowledge passing through matter, testing hypotheses against continuous material functioning reality; superior in forcing explicit principles articulation, synthesis unfunctioning, error obvious, demonstrated in matter. Third, ontologically, synthesis gesture living executes upon self, non-violation of external sacred/transcendent domain; creator belongs to creature category; continuity, non-rupture. Synthesis not merely technically viable (Section 2), epistemologically legitimate (technical knowledge theories); ontologically justified, heart of what living does when making living: reorganising, expanding, opening to new possibilities, transcending prior limits.

Synthesis continues what life always does, central statement with clarity, force: synthesis continues reorganising matter per constraints faced. Natural evolution selection of random variations, functioning in environment persist, reproduce; unfunctioning disappear (holders die without sufficient offspring). Directed synthesis selection of intentional variations, choosing characteristics, projecting mutations, cultivating organism lineages per anticipated, comprehended criteria. Difference selecting agency: evolution natural selection, environment pressure, fundamentally blind natural selection (no perspective, plan, merely differential reproduction of variants); synthesis human reflexive intention, planning, consequences comprehension. Both material operations upon living; both inside same biochemical constraints; both select, one un-knowing selection, other knowing. Synthesis genuine evolutionary continuation with reflexive agency, non-rupture with evolution, evolution become conscious of self via one mode.

Synthesis generates genuine knowledge, statement without qualifications: non-"knowledge application existing before synthesis"; new knowledge generation via material manipulation; overcoming revealed ignorance, failed synthesis reveals uncomprehended gaps; extending cognoscibility, synthesis forces explicit articulation of intuited/presumed; access to truths observation never permitted attaining, biological processes only engineering reveals, emergences arising reconfiguring components. Genuine intellectual gain, human knowing domain extension, living comprehension deepening.

Synthesis legitimation weakens not responsibility, radicalises deeply. Synthesis legitimate, responsibility not (theology suggested inadequately) "abstain action, responsible": abstinence non-moral virtue, responsibility evasion, refusal to act when action possible. Responsibility: "acting, responsible for continuous action effects". Demanding: no escape; no law prohibiting synthesis, absolving by abstinence; material effects cascade action engenders, propagating after initial action ends. Responsibility corresponds to cascade, vigilance over un-anticipatable effects.

Three vigilance domains synthesis demands: First, creature, synthesising suffering-capable being (suffering capacity feeling material constraints, experiencing inadequacy between needs and environment), responsible for well-being, world integration, continued living possibility; non-transcendent moral obligation from outside, , direct material consequence of creating sensing being: creating sentient being, immediate responsibility for well-being. Second, ecosystem, synthetic organism entering ecosystem (relationship, competition, predation, symbiosis network), responsible containing un-intentional cascade effects risks; organism invading ecological niches, outcompeting native species, altering nutrient fluxes, reconfiguring biogeochemical cycles, whole system structure; ecological vigilance material demand, non-moralism, material, real effects. Third, future, creating new possibility (new organism type, biological capacity, natural/technical life configuration), responsible comprehending precedents created; life synthesis today opens future possibilities un-anticipatable completely, new organism types, ecological relations, existing biota challenges, new opportunities for life.

Vigilance non-single completed act, non-"verified synthesis safe, proceed indefinitely without vigilance"; permanent attention process: continuous verification, revision per new knowledge, disposition to halt if consequences harmful. Vigilance, permanent attentive presence state to action-engendered effects cascade; metabolically incorporated: non-external law from outside, technical living possibility condition; synthesis damaging ecosystem, damages own living possibility conditions (depending on ecosystem).

Legitimation and radicalisation coexist in synthesis, non-contraries, two faces of same movement. Synthesis legitimate: living reorganising self reflexively, heart of life nature; continuation of operation life always executes, legitimation. Responsibility radical: synthesis unregulated by pre-existing transcendent law, , real material consequences, human capacity anticipating, responding to consequences. No transcendent foundation authorising/prohibiting synthesis, renders responsibility more demanding; continuous immanence, agents (natural, human, technical) operating contiguously, each operation engendering continuing, propagating, reverberating effects in un-completely knowable/predictable futures.

Life synthesis legitimate act, maximum responsibility act: legitimate, heart of living operation (reorganising); responsible, continuous vigilance obligation, attention to effects, comprehending un-anticipatable. No privilege, exposure; dependence relation with created, implication in generated effects; stating: "Synthesis legitimate, completely responsible for consequences."

Synthesis is the legitimate gesture of a living being possessing no fixed essence, because life is a regime that continues to exceed any definition we impose upon it, continuing to create what previously did not exist.

3.4. Closure: Reflexivity Without Foundation

Three movements, rigorous theological demolition, technical knowledge epistemological foundation, incorporated responsibility ethical radicalisation, converge to question chapter articulates forcefully, precisely, next chapter addresses deeply: if life possesses no fixed essence, synthesis gesture living executes upon self reflexively, responsibility incorporated in immanence without transcendence appeal, , regime defining living? Question posed comprehending what creating when creating life? Life nature exceeding prior definitions imposed?

Path, through chapter's three sections, revealed essential regarding synthesis: non-divine law transgression; operation life always executed, reorganising self, creating new possibilities, expanding domain over vast evolutionary time. Technical human no exception, most reflexive, conscious realisation of fundamental life capacity; non-human exceptional in essence, , life by nature reflection upon self, continuous capacity operating upon operation processes, reorganising matter made of per applied intelligence, intention to material constraints.

Means radical statement: life itself reflexivity; no pure life separate from life reflection; no nature preceding intelligence, interweaving of material processes and matter's capacity comprehending self; whole biosphere continuous feedback network where processes generate processes generating first. Life synthesis demonstration of reflexivity to conscious elaboration point, mechanisms comprehension permitting intentional modification.

Challenges Western philosophical thinking structuring categories for centuries: subject/object unsustained, synthesising observer living, part of observed; no outside position where knowledge objective, neutral, un-contaminated; inside life perspectives, always implied in observed. Nature/culture unseparated, known nature passes through intelligence, culture operation upon natural matter, deep inextricably. Essence/variation unopposed, essence continues through variation, life regime where continuous variation persistence condition, change constant, form provisional; no form fixed eternally, , temporary reorganisation of continuing processes.

Comprehension emerging from life synthesis: responsibility rests not on transcendent foundation; rests on radical immanence: material fact agent (human or non) implied in action-engendered consequences, material continuity between operation and effect, no separation between action and world unmade/remade by action. Life synthesis immanence demonstration: synthesising life, created life lives in same world inhabited; operations interwoven, effects affecting; no separation creator/creature (same shared immanence fabric).

Life synthesis fundamental philosophical question, non-merely technical: non-engineering problems (engineering, technical expertise resolve); forces un-postponable ontological question: living reorganising living reflexively, no creator/creature separation, all continuous immanence, , what is life? Regime persisting across transformations synthesis operates? Living nature radically reorganisable, genes modified, structure reconfigured, new capacities introduced, , remaining genuinely alive, metabolising, reproducing, responding to environment?

Next chapter addresses in systematic depth. Answer lies not in fixed essence discoverable archaeologically, ontologically, philosophically; lies in life comprehension as regime: continuous operation reorganising self, remaining through permanent transformation, opening unpredicted possibilities. Life continues exceeding prior imposed definitions. Synthesis radical indetermination proof, executed synthesis generates un-anticipated forms, capacities, opening new futures un-comprehensible prior to synthesis start, revealing comprehension incomplete, synthesis discovering un-predictable by observation.

Living regime continues when everything changes, non-preserved essence, capacity reorganising, remaining in operation. Synthesis discovers not regime, reveals operationally; evidences life no thing property, , thing operation. Operation continues in new forms, different substrates, unexisted configurations. Synthesis living proof of living matter fundamental freedom: capacity reorganising, creating continuously, un-determined by past.

The living is reflection without foundation, a continuous capacity to reorganise, know, and create itself, without appeal to a transcendent essence sustaining the transformation, as a regime that continues because it continues to operate.

Aphorisms:

  1. The human is a gesture, not a limit.
  2. Knowledge in biological engineering is not the application of prior theory, it is the generation of new truth through the act of making.
  3. The creator does not stand outside the creature, it is a creature become reflexive.
  4. The living is a regime, not an essence, a continuous operation that can be radically reorganised and still remain a living operation.
